Abstract
Profiling a full spectrum of biomarkers involved in inflammatory, autoimmune, and infectious disease remains a major challenge for diagnostic and therapeutic development, largely because existing multiplex platforms lack the sensitivity to capture proteins across a wide range of concentrations in blood. To meet this need, we developed the NULISAseq Immune 340 Panel, enabling simultaneous quantification of approximately 340 proteins spanning apoptosis, immune checkpoints, complement activation, and disease-associated pathways relevant to systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), inflammatory bowel disease (IBD), sepsis, and cancer.
The assay was optimized for plasma and serum and characterized for precision, reproducibility, detectability, specificity, and dilution linearity. Median intra- and inter-plate CVs were below 10%, over 90% of targets showed strong dilutional linearity (R² > 0.9) and >50% detectability with strong signal-to-noise ratio in plasma, and cross-reactivity remained under 1%. Applied to SLE serum and RA plasma cohorts, the panel revealed condition-specific signatures aligned with known disease biology, including elevated interferon and cell death/inflammasome markers in SLE, and higher levels of markers associated with TNF, IL-6/JAK-STAT, and Th17/IL-23 pathway activation in RA.
The NULISAseq™ Immune 340 Panel combines breadth, sensitivity, and reproducibility in a single assay, offering a powerful tool for immune pathway characterization, patient stratification, and early disease detection across diverse conditions.
